French election front-runner makes deficit pledge (AP)

PARIS ? The front-runner in France's presidential race vowed Thursday that if elected he'll bring the state's bloated deficit under the European Union's 3-percent target by next year by slashing tax breaks for corporations and the rich.

Socialist Francois Hollande said he was "in the same frame of mind" as U.S. President Barack Obama about the need to ask more of the wealthy at a time of hefty state-budget burdens and sluggish economic growth.

Laying out a 60-point program, Hollande said his tax plan could raise euro29 billion ($37.5 billion) in new income and help offset the cost of new initiatives to the tune of euro20 billion to support jobs, public housing and healthcare.

"Those who have benefited from crazy pay levels will have to make an effort," he said, promising to raise the tax bracket for the highest earners to 45 percent and to cap tax breaks for individuals at euro10,000 per year.

Hollande's appearance at a Paris news conference put meat on the bones' of his platform ? mostly on economic issues ? at a time when most polls show him with a comfortable lead over President Nicolas Sarkozy.

The conservative incumbent has not yet announced whether he will run for the two-round election in April and May, but nearly all political observers expect he will.

Hollande elaborated on a theme that he laid out at a campaign rally on Sunday, taking a pronounced leftist posture by announcing that his "real adversary in this campaign is the world of finance."

He also picked up a theme mentioned by Obama during his State of the Union address on Tuesday, when the U.S. leader spoke about how billionaire investor Warren Buffet pays a lower tax rate than Buffet's secretary.

"Obama said he wants the secretary of a billionaire to not have to pay more than the billionaire," Hollande said. "I want the same thing."

Hollande didn't mention Sarkozy by name, but implicitly targeted his tenure by citing a litany of gloomy French economic indicators and missed budget targets.

"And they want to give us lessons about budgetary responsibility?" Hollande said of Sarkozy's conservative government.

Hollande also slammed economic forecasts of the government's 1-percent growth target this year as too rosy. He predicts 0.5 percent growth ? much closer to independent forecasts from the IMF and World Bank.

The GOP, from Congress to the campaign trail, signaled it's ready for the fight.

In his third State of the Union address, Obama issued a populist call for income equality that echoed the Occupy Wall Street movement. He challenged GOP lawmakers to work with him or move aside so he could use the power of the presidency to produce results for an electorate uncertain whether he deserves another term.

Facing a deeply divided Congress, Obama appealed for lawmakers to send him legislation on immigration, clean energy and housing, knowing full well the election-year prospects are bleak but aware that polls show that the independent voters who lifted him to the presidency crave bipartisanship.

"I intend to fight obstruction with action," Obama told a packed chamber and tens of millions of Americans watching in prime time. House Republicans greeted his words with stony silence.

The Democratic president's vision of an activist government broke sharply with Republican demands for less government intervention to allow free enterprise. The stark differences will be evident in the White House's dealings with Congress and in the presidential campaign over the next 10 months.

In the Republican response to the president's address, Indiana Gov. Mitch Daniels, who once considered a White House bid, railed against the "extremism" of an administration that stifles economic growth.

"No feature of the Obama presidency has been sadder than its constant effort to divide us, to curry favor with some Americans by castigating others," Daniels said, speaking from Indianapolis. "As in previous moments of national danger, we Americans are all in the same boat."

Obama said getting a fair shot for all Americans is "the defining issue of our time." He described an economy on the rebound from the worst economic crisis since the Great Depression, with more than 3 million jobs created in the last 22 months and U.S. manufacturers hiring. Although unemployment is high at 8.5 percent, home sales and corporate earnings have increased, among other positive economic signs.

Republicans say the president's policies have undermined the economy.

Obama "had the opportunity and the responsibility to level with the American people, admit that the policies of the past three years have delivered an underwhelming record of economic growth and job creation, and show an interest in changing direction and uniting, not dividing the nation," said Rep. Tom Price, R-Ga., head of the Republican Policy Committee. "The president failed to meet that responsibility."

There were brief moments of bipartisanship. Republicans and Democrats sat together, continuing a practice begun last year. The arrival of Rep. Gabrielle Giffords, who survived an assassination attempt, elicited sustained applause and cheering, with chants of "Gabby, Gabby." Republican Rep. Jeff Flake escorted her into the chamber and Obama greeted her with a hug.

The president received loud applause from both sides when he said: "I'm a Democrat. But I believe what Republican Abraham Lincoln believed: That government should do for people only what they cannot do better by themselves, and no more."

But all that belied a fierce divide.

Obama ticked off items on a hefty agenda that he wants from Congress ? a path to citizenship for children who come to the United States with their undocumented parents if they complete college, tax credits for clean energy, elimination of red tape for Americans refinancing their mortgages, a measure that bans insider trading by lawmakers and a payroll tax cut.

Political reality suggests it was largely wishful thinking on Obama's part. The payroll tax cut and must-do spending bill are the most likely legislative items to survive the election year.

But Obama's far-reaching list and the hour-plus speech offered a unique opportunity to contrast his record with congressional Republicans and his top presidential rivals, Mitt Romney and Newt Gingrich.

"Anyone who tells you America is in decline or that our influence has waned, doesn't know what they're talking about," Obama said ? a clear response to the White House hopefuls who have pummeled him for months.

In an attack on the nation's growing income gap, Obama called for a new minimum tax rate of at least 30 percent on anyone making more than $1 million. Many millionaires ? including Romney ? pay a rate less than that because they get most of their income from investments, which are taxed at a lower rate.

"Now you can call this class warfare all you want," Obama said. "But asking a billionaire to pay at least as much as his secretary in taxes? Most Americans would call that common sense."

Obama calls this the "Buffett rule," named for billionaire Warren Buffett, who has said it's unfair that his secretary pays a higher tax rate than he does. Emphasizing the point, Buffett's secretary, Debbie Bosanek, attended the address in first lady Michelle Obama's box.

Obama made his appeal on the same day that Romney released some of his tax returns, showing he made more than $20 million in a single year and paid around 14 percent in taxes, largely because his wealth came from investments.

In advance of Obama's speech, Romney said, "Tonight will mark another chapter in the misguided policies of the last three years ? and the failed leadership of one man."

Obama highlighted his national security successes ? the raid that killed Osama bin Laden, the diminished strength of al-Qaida and the demise of Moammar Gadhafi. In hailing the men and women of the military, the commander in chief contrasted their cooperation and dedication with the divisions and acrimony in Washington.

"At a time when too many of our institutions have let us down, they exceed all expectations," Obama said. "They're not consumed with personal ambition. They don't obsess over their differences. They focus on the mission at hand. They work together. Imagine what we could accomplish if we followed their example."

Obama leaves Washington for a three-day tour of five states crucial to his re-election bid. On Wednesday he'll visit Iowa and Arizona to promote ideas to boost American manufacturing; on Thursday in Nevada and Colorado he'll discuss energy; and in Michigan on Friday he'll talk about college affordability, education and training.

He also addresses a conference of House Democrats focused on their own re-election in Cambridge, Md., on Friday.

Polling shows Americans are divided about Obama's overall job performance but unsatisfied with his handling of the economy.

Isner, the last American man left in the singles draw, lost to Lopez 6-3, 6-7 (3), 6-4, 6-7 (0), 6-1. It was the 6-foot-9 Isner's second straight five-set match after he knocked out David Nalbandian in a 4-hour, 41-minute thriller.

"It's very ugly, to be honest, to have no one in the round of 16," Isner said. "We've got to try to rectify that next time the big tournaments roll around.

"It's very disappointing. That's not a good effort from the Americans in this tournament. I knew going in today I was the last one left and I wanted to keep on going, but just didn't happen."

Instead, Lopez earned a fourth-round match against Rafael Nadal, who won the 2009 Australian Open and has 10 major titles to his credit.

"We've been spoiled with the success we've had in the past," McEnroe said.

The pressure of being last man standing got to Isner on Friday when he lapsed in the last set after dominating the fourth-set tiebreaker.

"It just got away from me. I just wasn't as sharp as I needed to be," he said. "It just kind of spiraled out of control there, and it just ... I couldn't climb out of the hole I dug."

"I honestly felt like it was more mental than anything," he added. "Just like the whole match I wasn't really that sharp."

Now he has to consider a Davis Cup series next month against a Swiss team that could feature Federer and Stanislas Wawrinka.

"That's not an ideal draw for a first round. But, yeah, it's going to be tough," Isner said. "It's away, and, you know, more than likely I think if Roger plays we're probably the underdogs."

Djokovic started 2011 on a 41-match winning streak and finished it with the No. 1 ranking and three of the four major titles. His two main rivals are already through to the third round on the other side of the draw.

The first suit was voluntarily dismissed. The second suit was concluded with the grant of summary judgment to the defendants.

The marriage ended in September 2007.

(AP) ? Records show the wife of Los Angeles Lakers guard Kobe Bryant will keep their three California homes as part of the couple's divorce plans.

Orange County property records reviewed by the Los Angeles Times (http://lat.ms/xUxzQv) show that the three mansions in Newport Coast, valued at $18.8 million total, were transferred into Vanessa Bryant's name since their divorce proceedings began in December.

Vanessa Bryant filed a divorce petition in Orange County Superior Court in December, citing "irreconcilable differences." The couple released a joint statement then saying that they had "resolved all issues incident to their divorce privately."

The Times says neither of the attorneys representing the couple was available for comment.

Elusive Z- DNA found on nucleosomes

New research published in BioMed Central's open access journal Cell & Bioscience is the first to show that left-handed Z-DNA, normally only found at sites where DNA is being copied, can also form on nucleosomes.

The structure of DNA which provides the blueprint for life has famously been described as a double helix. To save space inside the nucleus, DNA is tightly wound around proteins to form nucleosomes which are then further wound and compacted into chromatin, which is further compacted into chromosomes.

But this familiar image of a right handed coil (also called B-DNA) is not the only form of DNA. At sites where DNA is being copied into RNA (the messenger which is used as the instruction to make proteins) the DNA needs to unwind, and, in a process of negative supercoiling, can form a left-handed variety of the DNA double helix (Z-DNA).

It was originally thought that Z-DNA could only be formed in the presence of active RNA polymerase (the enzyme which assembles RNA). However more recently it has been discovered that SWI/SNF, a protein involved in remodeling nucleosomes and allowing RNA polymerase access to DNA, can convert certain sequences of B to Z-DNA.

The team of researchers led by Dr Keji Zhao discovered that they could convert B-DNA to Z-DNA on nucleosomes by the addition of SWI/SNF and ATP (the cell's energy source) and that the Z-nucleosome formed was a novel structure.

Dr Zhao, from the NIH, explained, "The fact that we have found Z-DNA on nucleosomes is a new step in understanding the roles of chromosome remodeling and Z-DNA in regulating gene expression. While the Z-nucleosome is likely to be a transient structure it nevertheless provides a window of opportunity for the placement of DNA binding proteins which may recruit, regulate, or block the transcription machinery and hence protein expression."
